摘要 | 为探究GATA转录因子在薄荷(Mentha canadensis Linn.)中的生物学功能,根据薄荷转录组数据克隆了McGATA5基因,并分析了McGATA5的序列结构、理化特性、系统进化关系、转录自激活活性,McGATA5的组织表达特性以及激素诱导和非生物胁迫条件下McGATA5在叶片中的表达水平变化。结果显示:薄荷McGATA5定位于细胞核,基因编码区(CDS)全长849 bp,编码282个氨基酸;McGATA5为亲水性蛋白,理论相对分子质量为30 931,理论等电点为pI 6.71。系统进化分析结果表明McGATA5属于GATA Ⅰ亚家族。转录自激活活性验证结果表明McGATA5具有转录自激活活性。组织表达分析结果显示McGATA5在薄荷根、茎、叶、花中均有表达,且在茎、成熟叶、花中的相对表达量显著高于根和幼叶。0.5 μmol·L-124-表油菜素内酯(EBR)、0.1 mmol·L-1脱落酸(ABA)、0.01 mmol·L-1萘乙酸(NAA)和200 mmol·L-1NaCl以及干旱(控水)处理后的McGATA5相对表达量明显上调,低温(4 ℃)处理后的McGATA5相对表达量总体下调。综上所述,薄荷McGATA5受激素EBR、ABA、NAA诱导表达,并参与NaCl、低温和干旱胁迫的应答过程。 |
Abstract | To investigate the biological functions of the GATA transcription factor in Mentha canadensis Linn., the McGATA5 gene was cloned based on the transcriptome data of M. canadensis, and the sequence structure, physicochemical properties, phylogenetic relationships, transcriptional self-activating activity of McGATA5, and tissue expression pattern of McGATA5, as well as variations in expression levels of McGATA5 in leaves under hormone induction and abiotic stress conditions were analyzed. The results show that McGATA5 of M. canadensis is localized in the nucleus, and the full length of coding sequence (CDS) is 846 bp, encoding 281 amino acids; McGATA5 is a hydrophilic protein, its theoretical relative molecular mass is 30 931, and its theoretical isoelectric point is pI 6.71. The phylogenetic analysis result indicates that McGATA5 belongs to subfamily Ⅰ of GATA. The transcriptional selfactivating activity verification result shows that McGATA5 possesses transcriptional self-activating activity. The tissue expression analysis result reveals that McGATA5 is expressed in roots, stems, leaves, and flowers of M. canadensis, and the relative expression in stems, mature leaves, and flowers are significantly higher than those in roots and young leaves. The relative expression of McGATA5 are evidently up-regulated after treated with 0.5 μmol·L-1 24-epicastasterone (EBR), 0.1 mmol·L-1 abscisic acid (ABA), 0.01 mmol·L-1 naphthaleneacetic acid (NAA), and 200 mmol·L-1 NaCl, as well as under drought (water control) treatment, while the relative expression of McGATA5 is generally down-regulated after low-temperature (4 ℃) treatment. In conclusion, the expression of McGATA5 in M. canadensis is induced by the hormones EBR, ABA, and NAA, and it is involved in the response processes to NaCl, low temperature, and drought stress.
