猴头子实体锰型超氧物歧化酶的纯化及其性质鉴定

摘要

猴头(Hericium erinaceus(Bull.)Pers.)子实体的超氧物歧化酶仅Mn-SOD1种,有其资源和学术研究上的意义。其粗酶液经(NH4)2SO4盐析,DE52和CM52离子交换柱层析,纯化到电泳单斑点均一程度,其比活性为3096.5u/mg,活性回收率为14.8%。纯化的Mn-SOD分子量为47.0KD,亚基分子量为23.3KD。金属元素分析表明每个亚基含1个Mn原子。该酶在紫外区有2个吸收峰,分别在218.4um和276.0um。该酶的理化性质与文献报道的不同来源的Mn-SOD大致相同。 

Abstract

Only one kind of Mn-superoxide dismutase (Mn-SOD) was found in the crude extract from Hericiurn erinaceus (Bull. ) Pers. In this case, there are significance of resources and scientific study. The enzyme was isolated and purified to homogenity by ammonium sulfate fraction, DE52 and CM52 chromotography ,and the rate of activity yield is 18. 8 % . The enzyme had a specific activity of 3 096.5 μ/mg, and exhibited two obsorption peaks in the ultraviolet at 218.4 nm and 276.0 nm respectively. The purified enzyme Mn-SOD has a relative molecular mass of 47.0 KD and the molecular mass of subunit is 23.3 KD. Analysis of metalic elements demonstrated that every subunit contains one Mn atom. The physiochemical properties of the enzyme are roughly the same with the same kind of SOD from different sources reported by literatures.
