渗透胁迫条件下内生细菌对银砂槐种子萌发和主要碳水化合物含量的影响
Effects of endophytic bacteria on seed germination and main carbohydrate contents in Ammodendron bifolium under osmotic stress condition

摘要

为了探明渗透胁迫条件下内生细菌对银砂槐〔Ammodendron bifolium (Pall.) Yakovl.〕种子萌发和早期幼苗生长的影响,在正常(渗透势0.0 MPa)和PEG6000模拟的渗透胁迫(渗透势-0.3 MPa)条件下,对培养0、5、10和15 d接菌组(分别接种内生细菌菌株AY3、AY9和AG18,即AY3、AY9和AG18组)和对照(未接种内生细菌,CK)组银砂槐的种子萌发(种子萌发率和胚根长)及主要碳水化合物(包括淀粉、可溶性糖、蔗糖、葡萄糖、果糖和总碳水化合物)含量进行了比较分析。结果表明:总体上看,渗透胁迫条件下接菌组和CK组的种子萌发率、胚根长及葡萄糖和果糖含量均低于正常条件下,而淀粉、可溶性糖、蔗糖和总碳水化合物含量均高于正常条件下;并且,正常和渗透胁迫条件下接菌组的种子萌发率、胚根长及葡萄糖和果糖含量均高于CK组,而淀粉、可溶性糖、蔗糖和总碳水化合物含量均低于CK组。另外,正常和渗透胁迫条件下接菌组和CK组的葡萄糖含量在培养5 d时均为0.00 mg·g-1。研究结果显示:渗透胁迫能够抑制银砂槐种子萌发和胚根生长;内生细菌可明显减轻渗透胁迫对银砂槐种子萌发和胚根生长的抑制效应,并能促进其种子萌发过程中碳水化合物的动员和利用。

 

Abstract

In order to explore the effects of endophytic bacteria on seed germination and early seedling growth of Ammodendron bifolium (Pall.) Yakovl. under osmotic stress condition, seed germination (including seed germination rate and radicle length) and main carbohydrate (including starch, soluble sugar, sucrose, glucose, fructose and total carbohydrate) contents in A. bifolium in inoculation groups (inoculated with endophytic bacterium strain AY3, AY9 and AG18, respectively, namely AY3, AY9 and AG18 groups) and the control (not inoculated with endophytic bacterium, CK) group cultured for 0, 5, 10 and 15 d were compared and analyzed under normal (osmotic potential of 0.0 MPa) and osmotic stress (osmotic potential of -0.3 MPa) simulated by PEG6000 conditions. The results show that on the whole, seed germination rate, radicle length, and glucose and fructose contents in inoculation groups and CK group under osmotic stress condition are all lower than those under normal condition, while starch, soluble sugar, sucrose and total carbohydrate contents are all higher than those under normal condition. In addition, seed germination rate, radicle length, and glucose and fructose contents in inoculation groups under normal and osmotic stress conditions are all higher than those in CK group, while starch, soluble sugar, sucrose and total carbohydrate contents are all lower than those in CK group. Besides, when cultured for 5 d, glucose content in inoculation groups and CK group is 0.00 mg·g-1 under normal and osmotic stress conditions. It is suggested that osmotic stress can inhibit seed germination and radicle growth of A. bifolium; endophytic bacteria can obviously alleviate inhibition effects of osmotic stress on seed germination and radicle growth of A. bifolium, and can improve mobilization and utilization of carbohydrate during its seed germination process.
