摘要 | 利用超高效液相色谱四极杆飞行时间串联质谱联用(UPLC-Q-TOF-MS)代谢组学技术,对地涌金莲〔Musella lasiocarpa (Franch.) C. Y. Wu ex H. W. Li〕等6种芭蕉亚科(subfam. Musoideae)植物种子代谢物进行非靶向分析,并进一步对地涌金莲不同成熟期种子及不同组织的代谢物进行靶向分析,还对7种化合物进行了体外抗真菌活性检测。结果显示:地涌金莲种子代谢物与芭蕉亚科另5种植物差异明显。地涌金莲与蕉麻(Musa textilis Née)之间的显著差异代谢物最少(279),与朝天蕉(Musa velutina H. Wendl. et Drude)之间的显著差异代谢物最多(429);地涌金莲与另5种植物的共有显著差异代谢物数量为63,识别鉴定出37个代谢物,其中,4个二芳基庚烷类化合物在地涌金莲种子中的相对含量显著高于另5种植物。靶向分析结果显示:在地涌金莲4个成熟期种子中共鉴定到12个二芳基庚烷类显著差异代谢物,且多数化合物在黑色或深褐色种子中的相对含量最高。对地涌金莲不同组织中的二芳基庚烷类化合物含量进行比较,结果显示这类化合物多集中在地下组织(假茎、假茎皮、根)。抗真菌活性实验结果显示:2-methoxy-9-phenyl-1H-phenalen-1-one对尖孢镰刀菌(Fusarium oxysporum)、胶孢炭疽菌(Colletotrichum gloeosporioides)和链格孢菌(Alternaria alternata)的抑制率显著(P<0.05)高于其他化合物。综合分析结果显示:地涌金莲富含二芳基庚烷类化合物,且多数二芳基庚烷类化合物为苯基非那烯酮类化合物,且具有较高的抗真菌活性。

Abstract | Through using ultra performance liquid chromatographyquadrupoletime of flight-mass spectrometry (UPLC-Q-TOF-MS) metabolomics technology, non-targeted analysis was conducted for the metabolites in seeds of six subfam. Musoideae species such as Musella lasiocarpa (Franch.) C. Y. Wu ex H. W. Li, and targeted analysis was further performed for the metabolites in seeds at different maturation stages and different tissues of M. lasiocarpa, in vitro antifungal activity was also conducted for seven compounds. The results show that the metabolites in M. lasiocarpa seeds are evidently different from those in the other five Musoideae species. The significantly differential metabolites between M. lasiocarpa and Musa textilis Née are the fewest (279), while those between M. lasiocarpa and M. velutina H. Wendl. et Drude are the most (429); the number of common significantly differential metabolites between M. lasiocarpa and the other five species is 63, with 37 metabolites identified, among which, the relative contents of four diarylheptanoids compounds in M. lasiocarpa seeds are significantly higher than those in the other five species. The targeted analysis result shows that 12 significantly differential diarylheptanoids metabolites are identified in seeds of M. lasiocarpa at four maturation stages, and the relative contents of most compounds are the highest in black or dark brown seeds. The contents of diarylheptanoids compounds in different tissues of M. lasiocarpa were compared, and the result shows that these compounds are mostly concentrated in the underground tissues (pseudostems, pseudostem barks, and roots). The antifungal activity assay result shows that the inhibition rate of 2-methoxy-9-phenyl-1H-phenalen-1-one against Fusarium oxysporum, Colletotrichum gloeosporioides, and Alternaria alternata is significantly (P<0.05) higher than that of the other compounds. The comprehensive analysis result indicates that M. lasiocarpa is rich in diarylheptanoids, and most of them are phenylphenalenones with relatively high antifungal activity.
