In the tissue culture only the calli from axillary buds of Lactuca sativa L .var.yemena X.S .Shen et X .P.Zhang can be differentiated into seedlings on the designed medium, although both leaves and buds are successful for callus induction .The combination of NAA 0 .5 mg/ L and BA 3.0 mg/ L as an induced medium is optimum for the formation of the calli, whereas in bud regenerating medium, the concentration of NAA has to be reduced from 0.5 to 0.2 mg/ L, which is suitable for regeneration of buds. The growth of test-tube plantlets and the effects of certain related factors are discussed .
