急性子中 balsaminone A 和 balsaminone B 对人肺癌 A549 细胞生长及周期的影响

摘要

应用 MTT 法和流式细胞仪研究了急性子(Impatiens balsamina L.)中双萘呋喃-7,12-酮类化合物 balsaminoneA 和 balsaminone B 对人肺癌 A549 细胞生长及周期的影响。 结果表明:20、40、60 和 80 μmol· L-1 balsaminone A 和B 作用 48 h 均对 A549 细胞生长的抑制作用显著(P<0. 05),其吸光值均显著低于阴性对照且随浓度的提高逐渐降低,但均高于阳性对照(100 mg· L-15-氟尿嘧啶); 各处理组 A549 细胞的生长抑制率均低于阳性对照组, 其中60 和 80 μmol· L-1 balsaminone A 处理组的生长抑制率接近阳性对照。 balsaminone A 作用 48 h 使 DNA 合成前期(G0 / G1期)的细胞比例减少,DNA 合成期(S 期)和 DNA 合成后期(G2 / M 期)的细胞比例增加,但无明显的量效关系;而 balsaminone B 作用 48 h 使 G0 / G1期的细胞比例减少,S 期的细胞比例明显增加,呈明显的量效关系。 研究结果显示:balsaminone A 和 B 对 A549 细胞的生长均具有显著的抑制作用,二者的作用机制可能与诱导细胞周期阻滞有关但有一定的差异。

Abstract

The effects of dinaphthofuran-7, 12-dione derivatives, which are balsaminone A and balsaminone B, from Impatiens balsamina L. seed on growth and cycle of human lung cancer A549 cell were studied by MTT assay and flow cytometry. The results show that both balsaminone A and B with 20, 40, 60 and 80 μmol· L-1 have significant inhibition effects on growth of A549 cell after 48 h (P< 0. 05), and their absorption values, which gradually decrease with enhancing of concentrations, all are lower than those of the negative control but higher than those of the positive control (100 mg· L-1 5-FU). The growth inhibition rate of A549 cell in all treatment groups is lower than that of the positive control group, in which, that in 60 and 80 μmol· L-1 balsaminone A treatment groups is close to that of the positive control. Balsaminone A leads to decreasing of cell proportion in DNA presynthesis phase (G0 / G1 phase) and to increasing of cell proportion in DNA synthesis phase ( S phase) and DNA postsynthesis phase ( G2 / M phase ) after 48 h with an unobvious dose-effect relationship. While balsaminone B leads to decreasing of cell proportion in G0 / G1 phase and to evidently increasing of cell proportion in S phase after 48 h with an obvious dose-effect relationship. It is suggested that balsaminone A and B have significant inhibition effects on growth of A549 cell, their action mechanism may be related to induction of cell cycle arrest but their effects exist some differences.
