摘要 | 用新鲜的或-75 ℃保存的苍术〔Atractylodes lancea ( Thunb .) DC .〕叶片提取 DNA, 产量分别为 40 ng/ mg 鲜重和 10 ng/mg 鲜重左右, 分离出的 DNA 的 OD260/ OD280值均大于 1.9, 可用于RA PD 试验。 用 8 种引物对苍术 4 个居群的 DNA 进行扩增, 单个 10 碱基的引物扩增出的 RAPD标记在1 ~ 15 之间, 多态位点百分率南苍术为 47.50%, 北苍术为 45.40%, 遗传相似度值南苍术为0.85, 北苍术为 0.87 。 结果表明, 南苍术的遗传多样性略高于北苍术。 |
Abstract | The DNA of fresh or stored ( under -75 ℃) leaves of Atractylodes lancea ( Thunb .) DC .were isolated, the yield is about 40 ng/mg FW and 10 ng/mg FW separately .The OD260/OD280 values of isolated DNA are all above 1 .9, the DNA can be used in the test of RAPD .The amplified results of 4 populations by 8 primers indicated that single 10-base oligonucleotide primer can produce 1 to 15 pieces of DNA fragments .The percentage of polymorphic loci are 47 .50 % in south A .lancea and 45 .40 %in the north ones, the genetic similarity value is 0 .85 in the former and 0 .87 in the later, it showed that the genetic diversity level in the south A . lancea is slightly higher . |
