摘要 | 为探明茅苍术〔Atractylodes lancea (Thunb.) DC.〕种子的内源抑制物,采用系统溶剂分离法对茅苍术种子甲醇浸提液进行分离,共得到石油醚相、二氯甲烷相、乙酸乙酯相、甲醇相和水相5个分离相;以小白菜品种‘矮脚黄’(Brassica campestris ssp. chinensis var. communis ‘Aijiaohuang’)种子为受试对象,对茅苍术种子甲醇浸提液、不同分离相及6种可能内源抑制物进行生物活性检测。结果表明:在茅苍术种子甲醇浸提液及二氯甲烷相、乙酸乙酯相和甲醇相处理下,‘矮脚黄’的种子发芽率和胚根长度均显著(P<0.05)低于对照组(超纯水)。从茅苍术种子甲醇浸提液的石油醚相、二氯甲烷相、乙酸乙酯相和甲醇相中共分离鉴定出37种化合物,其中,酯类、酚类、有机酸类、醇类、醛酮类、苯类、甘油类、醚类和烷类分别有8、6、6、5、4、3、2、2和1种。在1 000 μmol·L-1邻苯二甲酸二丁酯、2-甲氧基-4-乙烯基苯酚、(2S,3R,4S,5S)-2-甲氧基四氢-2H-吡喃-3,4,5-三醇、(9E,11E)-共轭亚油酸、棕榈酸和硬脂酸处理下,‘矮脚黄’的胚根长度显著低于对照组,其中,邻苯二甲酸二丁酯和2-甲氧基-4-乙烯基苯酚处理组‘矮脚黄’胚根长度的抑制率分别为50.59%和60.81%,半抑制浓度分别为623.09和136.01 μmol·L-1。综上所述,邻苯二甲酸二丁酯和2-甲氧基-4-乙烯基苯酚可能是导致茅苍术种子发芽率低的主要内源抑制物。

Abstract | To explore the endogenous inhibitors in Atractylodes lancea (Thunb.) DC. seeds, the methanol extracts of A. lancea seeds were separated by using the systematic solvent partitioning method, and a total of five separation phases were obtained, namely petroleum ether phase, dichloromethane phase, ethyl acetate phase, methanol phase, and aqueous phase; taking the seeds of Brassica campestris ssp. chinensis var. communis ‘Aijiaohuang’ as the test subjects, biological activity detections were conducted for the methanol extracts of A. lancea seeds, different separation phases, and six potential endogenous inhibitors. The results show that the seed germination rate and radicle length of ‘Aijiaohuang’ under treatments of methanol extracts of A. lancea seeds, dichloromethane phase, ethyl acetate phase, and methanol phase are significantly (P<0.05) lower than those of the control group (ultrapure water). A total of 37 compounds are isolated and identified from the petroleum ether phase, dichloromethane phase, ethyl acetate phase, and methanol phase from the methanol extracts of A. lancea seeds, among which, esters, phenols, organic acids, alcohols, aldehydes and ketones, benzenes, glycerins, ethers, and alkanes are 8, 6, 6, 5, 4, 3, 2, 2, and 1 types, respectively. The radicle length of ‘Aijiaohuang’ under treatments of 1 000 μmol·L-1 dibutyl phthalate, 2-methoxy-4-vinylphenol, (2S,3R,4S,5S)-2-methoxytetrahydro-2H-pyran-3,4,5-triol, (9E,11E)-conjugated linoleic acid, palmitic acid, and octadecanoic acid is significantly lower than that of the control group, among which, the inhibitory rates of radicle length of ‘Aijiaohuang’ of dibutyl phthalate and 2-methoxy-4-vinylphenol treatment groups are 50.59% and 60.81%, respectively, and the half inhibitory concentrations are 623.09 and 136.01 μmol·L-1, respectively. In conclusion, dibutyl phthalate and 2-methoxy-4-vinylphenol may be the primary endogenous inhibitors causing low germination rate of A. lancea seeds.
