三种抗生素对丰水梨叶片不定芽再生的影响及其对根癌农杆菌的抑菌效果
Effects of three antibiotics on regeneration of adventitious buds from Pyrus pyrifolia `Hosui' leaf and inhibition to Agrobacterium tumefaciens

摘要

以丰水梨(Pyrus pyrifolia‘Hosui’)离体叶片为实验材料,研究了氨苄青霉素(Amp)、羧苄青霉素(Carb)和头孢霉素(Cef)对丰水梨叶片不定芽再生的影响及其对根癌农杆菌(Agrobacterium tumefaciens)的抑菌效果。结果表明,在100~500 mg.L-1浓度范围内,3种抗生素对丰水梨叶片愈伤组织诱导均无显著影响。Amp、Carb和Cef抑制丰水梨叶片不定芽再生的起始浓度分别为100、200和200 mg.L-1;当浓度高达500 mg.L-1时,Amp、Carb和Cef均极显著地抑制叶片不定芽再生。除500 mg.L-1Cef处理组外,不同浓度Amp、Carb和Cef对丰水梨每个外植体的再生不定芽数没有显著影响。与对照相比,在100~500 mg.L-1浓度范围内,3种抗生素均对根癌农杆菌菌株LBA4404和EHA105有显著的抑制作用,且对菌株EHA105的抑制效果强于菌株LBA4404。研究结果显示,在丰水梨遗传转化体系中,可以选择300 mg.L-1的Amp和Cef作为理想的杀菌剂。 

Abstract

Effects of ampicillin, carbenicillin and cefotaxime on regeneration of adventitious buds from Pyrus pyrifolia `Hosui' leaf and inhibition of the three antibiotics to Agrobacterium tumefaciens were studied within vitro leaves of P. pyrifolia `Hosui'. The results show that the three antibiotics (100- 500 mg·L- 1)have no significant effect on callus induction of the leaves. Adventitious bud regeneration starts to be restrainedat 100mg·L- 1of ampicillin and 200mg·L- 1of carbenicillin or cefotaxime. When the concentrations of the three antibiotics are up to 500mg·L-1, there generation of adventitious buds are extremely significantly inhibited. Ampicillin, carbenicillin and cefotaxime with different concentrations have no significant effects on the regenerated adventitious bud number per explant except 500mg·L-1 cefotaxime. Contrast to the control, reproduction of A. tumefaciens LBA 4404 and EHA 105 strains is significantly inhibited by the three antibiotics at 100-500mg·L- 1, and the inhibition to EHA 105 is better than that to LBA 4404. It is concluded that300mg·L- 1ampicillin and cefotaxime are ideal bactericides for P. pyrifolia `Hosui' genetictrans  formation system.
