Abstract | Foreign NPT Ⅱ gene was introduced into calli and mature embryos of wheat strains 922-267, 942-114 and 953-1575 by using PDS-1000/ He particle delivery system .Plasmid pZFX2 contains the NPT Ⅱ ( neomycin phosphotransferase) and gly colate oxidase gene which can increase the w heat plant resistance to insects .A fter selection culture based on MS medium containing 100 μg/mL Kanamycin, resistant calli w ere selected. Green plants were regenerated from part of resistant calli.Through this test, we found that the optimum pressure of bombarding calli was 1 100 psi.U sing this pressure, more resistant calli were obtained. Mature embryos were bombared at pressure of 1 300 psi.I t is w orth to notice that the green plants derived from transformed mature embry os were easier than those from transformed calli . |
