不同品种桑椹乙醇提取物的化学成分含量差异及抗卵巢功能衰退活性分析
Analysis on content differences of chemical components and anti-ovarian failure activity of ethanol extracts of mulberry of different cultivars

摘要

了初步明确桑椹的化学成分及抗卵巢衰退活性,对9个品种桑椹乙醇提取物的总多酚、多糖、花色苷、蛋白质、绿原酸及芦丁含量进行了检测和比较,并对其乙醇提取物的抗卵巢衰退活性进行了比较分析。结果显示:9个品种桑椹乙醇提取物的总多酚、多糖、花色苷、蛋白质、绿原酸和芦丁含量分别为8.60~18.05 、12.06~20.17、0.63~1.77、0.44~1.11、0.09~0.18和0.14~0.29 mg·g-1,6种化学成分含量的品种间差异极显著(P<0.01),其中,‘金樯63’(‘Jinqiang 63’)和‘嘉陵30’(‘Jialing 30’)的6种化学成分含量多显著(P<0.05)高于其他品种。与模型对照组相比,9个样品组的雌二醇和孕酮含量多显著升高,半胱天冬酶-3(Caspase-3)和B淋巴细胞瘤-2(Bcl-2)均下降,而Bcl-2相关X蛋白(Bax)含量和抗凋亡指数变化不显著,总体来看,‘金樯63’和‘嘉陵30’处理组表现出的卵巢活性恢复效果较好。综上所述,9个品种桑椹乙醇提取物的化学成分含量差异极显著并具有明显的抗卵巢衰退活性,尤其是‘金樯63’和‘嘉陵30’。

Abstract

To preliminarily clarify the chemical components and anti-ovarian failure activity of mulberry, the contents of total polyphenol, polysaccharide, anthocyanin, protein, chlorogenic acid, and rutin in ethanol extracts of mulberry of nine cultivars were measured and compared, and the anti-ovarian failure activity of their ethanol extracts was compared and analyzed. The results show that the contents of total polyphenol, polysaccharide, anthocyanin, protein, chlorogenic acid, and rutin in ethanol extracts of mulberry of nine cultivars are 8.60-18.05, 12.06-20.17, 0.63-1.77, 0.44-1.11, 0.09-0.18, and 0.14-0.29 mg·g-1, respectively, and there are extremely significant differences (P<0.01) in contents of these six chemical components between cultivars, in which, the contents of these six chemical components in ‘Jinqiang 63’ and ‘Jialing 30’ are mostly significantly (P<0.05) higher than those in the other cultivars. Compared with the model control group, the contents of estradiol and progesterone in the nine sample groups significantly increase in general, the contents of cysteinyl aspartate specific proteinase-3 (Caspase-3) and B-cell lymphoma-2 (Bcl-2) decrease, while the content of Bcl-2 associated X protein (Bax) and the index of anti-apoptosis show no significant variations, overall, the ‘Jinqiang 63’ and ‘Jialing 30’ treatment groups exhibit better recovery effects on ovarian activity. In conclusion, the chemical components of ethanol extracts of mulberry of the nine cultivars show extremely significant differences and exhibit evident anti-ovarian failure activity, especially in ‘Jinqiang 63’ and ‘Jialing 30’.
