摘要 | 采用两步培养法培养新疆紫(Arnebia euchroma (Royle) Johnst)草细胞以生产紫草色素,建立了从250ml摇瓶放大到9L和25L内循环气升式反应器的逐级放大培养程序。第一步细胞生长迅速,并有少量色素合成;第二步培养以生产色素为主,细胞生长明显减缓。研究了摇瓶培养时连续继代培养对细胞生长及色素合成的影响,以选择继代培养2~3代的细胞接入反应器中培养效果较好。9L反应器培养中通气率为0.4vvm时可得到较高色素产量,对25L反应器中混合及溶氧对细胞生长及色素合成的影响也进行了讨论。 |
Abstract | A two-stage culture method was developed to culture Arnebia euchroma (Royle) Johnst cells,and established a process of progressive scale-up culture from 250 ml shake flask to 9 L and 25 L airlift bioreactors with concentric draft tube. In the first stage, cells grew very fast, with the formation of a small amount of shikonin derivatives. In the second stage, cell growth was relatively slow, and a large amount of shikonin derivatives was produced. The influence of successive subculture in shake flask and different aeration rate of 9 L bioreactor on cell growth and shikonin derivatives formation was examined. The problem of mixing and oxygen supply on cell growth and product formation in 25 L bioreactor were also discussed. |
