香水莲花花蕊醇提物对黄嘌呤氧化酶的抑制作用及其主要成分

摘要

 通过体外实验测定了香水莲花(Nymphaea sp.)花蕊醇提物对黄嘌呤氧化酶(XOD)的抑制率及其动力学常数(Ki),并采用液质联用法分析该醇提物的主要成分。结果表明:香水莲花花蕊醇提物对XOD具有抑制作用,且随醇提物质量浓度的升高抑制率增大,半数抑制浓度(IC50)为38.54 μg·mL-1;抑制反应类型为混合型抑制,Ki值为0.95 mg·mL-1。该醇提物的主要成分为没食子酸、鞣花酸和槲皮苷,其中鞣花酸对XOD有一定的抑制作用(IC50值为7.56 μg·mL-1),而没食子酸和槲皮苷对XOD无抑制作用。

Abstract

The inhibitory rate of ethanol extract from stamen and pistil of Nymphaea sp. on xanthine oxidase (XOD) and its kinetic constant(Ki) were determined in vitro, and the main components of the ethanol extract were analyzed by using HPLC-MS method. The results show that the ethanol extract from stamen and pistil of Nymphaea sp. has inhibitory effect on XOD, the inhibition rate increases with the increase of ethanol extract mass concentration, and the half inhibitory concentration (IC50) is 38.54 μg·mL-1; the type of inhibition is mixed inhibition, and the Ki value is 0.95 mg·mL-1. The main components of the ethanol extract are gallic acid, ellagic acid, and quercitrin, in which, ellagic acid has a centain inhibitory effect on XOD (IC50 value is 7.56 μg·mL-1), but gallic acid and quercetin have no inhibitory effect on XOD.
