细胞分裂素种类及添加量对蓝浆果叶片离体再生的影响
Effects of type and adding amount of cytokinins on regeneration in vitro leaves of blueberries (Vaccinium spp.)

摘要

以南方高丛蓝浆果(Vaccinium corymbosum hybrids)品种‘南月’(‘Southmoon’)优选系A18 和兔眼蓝浆果(V. ashei Reade)品种‘灿烂’(‘Brightwell’)离体叶片为外植体, 研究了培养基中添加不同浓度TDZ(0. 5、1. 0 和2. 0mg·L-1 )、CPPU(2. 0、4. 0 和8. 0 mg·L-1 )、ZT(2. 0、4. 0 和8. 0 mg·L-1)和2iP(4. 0、8. 0 和16. 0 mg·L-1 )对叶片不定芽再生的影响。结果表明:在培养基中添加TDZ 和CPPU 对叶片不定芽的诱导效果优于ZT 和2iP,再生率有显著差异(P<0. 05)。TDZ 诱导不定芽出现所需的时间最短且再生率最高,不定芽密集并呈深绿色;其中,在添加0. 5 ~2. 0 mg·L-1TDZ 的培养基上,A18 叶片再生率均为100. 00%,‘灿烂’叶片再生率最高达79. 17%。CPPU 也有较强的诱导能力但不定芽出现所需的时间推迟3 ~5 d,且不定芽的密集程度也有所降低;其中,在添加2. 0 ~8. 0 mg·L-1CPPU 的培养基上,A18 叶片再生率为100. 00% ~93. 75%;而‘灿烂’叶片再生率随CPPU 质量浓度提高呈下降趋势(72. 91% ~47. 91%)。ZT 和2iP 诱导能力差,在添加不同质量浓度ZT 和2iP 的培养基上A18 和‘灿烂’叶片再生率均为0. 00%。此外,A18 和‘灿烂’的再生能力有差异,在相同条件下A18 叶片的再生能力优于‘灿烂’叶片。研究结果显示:基因型以及培养基中细胞分裂素的种类和添加量是影响不同品种蓝浆果叶片不定芽再生的主要因素。

Abstract

Taking in vitro leaves of superior strain A18 of cultivar ‘Southmoon’of Vaccinium corymbosum hybrids and cultivar ‘Brightwell’of V. ashei Reade as the explants, effect of different concentrations of TDZ (0. 5, 1. 0 and 2 . 0 mg·L-1), CPPU (2. 0, 4. 0 and 8. 0 mg·L-1), ZT (2. 0, 4. 0 and 8. 0 mg·L-1) and 2iP (4. 0, 8. 0 and 16. 0 mg·L-1) added in media on adventitious bud regeneration was studied. The results show that induction effect of adding TDZ and CPPU in media on adventitious bud is better than those of adding ZT and 2iP with the significant difference (P<0. 05) in regeneration rate. Required time of adventitious bud appearance induced by TDZ is the shortest and regeneration rate is the  highest, and adventitious buds are dense and dark green, in which, regeneration rate of A18 leaves all is 100. 00% and that of ‘Brightwell’is up to 79. 17% in media adding 0. 5- 2. 0 mg·L-1 TDZ. Also, CPPU possesses the relative strong induction ability but required time of adventitious bud appearance postpones 3-5 d, and adventitious bud intensity decreases, in which in media adding 2. 0-8. 0 mg·L-1 CPPU, regeneration rate of A18 leaves is 100. 00% -93. 75% and that of ‘Brightwell’ appears the decreasing trend (72. 91% -47. 91%) with rising of CPPU concentrations. Induction effect of ZT and 2iP is very poor, and regeneration rate of leaves of A18 and ‘Brightwell’all is 0. 00% in media adding different concentrations of ZT and 2iP. Otherwise, the regeneration ability is various between A18 and ‘Brightwell’, that of A18 leaf is superior to that of ‘Brightwell’leaf under same culture condition. It is suggested that genotype and amount and concentration of cytokinins added in media are main factors influencing on regeneration of adventitious buds of leaves of different cultivars of blueberry.
