Abstract | Taking stem tips of Iris foetidissima Linn. as explants, effect of different phytohormone combinations on its callus and adventitious bud inductions and effect of different basic media and mass concentrations of NAA on rooting were studied, the high frequency in vitro regeneration condition of I. foetidissima was selected preliminarily. Results show that the optimum callus induction medium of I. foetidissima is MS medium containing 1. 5 mg·L-1 2,4-D and 0. 1 mg·L-1 6-BA, its callus induction rate is high to 77. 27%. The optimum adventitious bud induction medium is MS medium added with 1. 0 mg·L-1 6-BA and 0. 2 mg·L-1 NAA, with adventitious bud induction rate of 72. 22%. The optimum rooting medium is MS medium added with 0. 2 mg·L-1 NAA, its rooting rate reaches to 100. 00%. The survival rate of plantlets after transplanted is also high with a value of above 95%. |
